Physiological condensates, pathological aggregates and low complexity sequences (LCS): Exploring the uncharted territory

Abstract: Our group aspires to address the molecular basis of mRNA fate decisions in the cytoplasm. We utilize RNA-binding proteins with low complexity sequences (LCS such as RGG motifs) as tools to understand the movements of mRNA in and out of translation. RNA granules or physiological condensates such as P-bodies stress granules play a key role in determining cytoplasmic mRNA fate at the level of mRNA translation and stability. LCS have been demonstrated to be important for the assembly of physiological condensates by promoting phase separation behavior of key residentproteins. However, we are currently exploring the role of these sequences in disassembly of physiological condensates. As an extension of the hypothesis, we are also testing the role of these sequences in the disassembly of pathological aggregates implicated in neurodegenerative disorders such as ALS. The recent results obtained using a combination of genetic, biochemical and live cell imaging approaches will be discussed.
